We are currently evaluating Design Studio 1.6. As datasource, we want to use BO universes (no SAP BW or SAP HANA). Is it possible to use unv-universes ?

The HELP Manual (help.SAP.com) for DesignStudio specifically mentions IDT when talking about universes, this leads me to assume only unX!
Also, remember that you must be running SAP BI BusObjects 4.1 or higher for DesignStudio to be able to access universes (unX) as a data source.
